HIV vaccines: immune targets may be key to a more effective preventative vaccine

Merck & Co has announced that it will be trialing a trivalent HIV vaccine in a Phase II proof-of-concept study with the US HIV Trials Network (HVTN). Despite nearly two decades of investigation, an effective HIV vaccine seems no nearer fruition. However, as Datamonitor's Laura Harris explains, more sophisticated immune targeting may yet bring progress towards at least a partially effective drug...

Eli Lilly: losing the war in antipsychotics

Eli Lilly [LLY] has announced an increase in revenues during Q4 2004 and a 10% increase in full year sales. However, revenues of its antipsychotic Zyprexa (olanzapine) fell significantly. A looming court decision on a US patent challenge to the drug further increases the uncertainty of Zyprexa's future. Even if Lilly wins in the courts, it is losing the battle in the antipsychotic market.

Sanofi-Aventis: Eloxatin consolidates its position of strength

Sanofi-Aventis has announced that a new formulation for Eloxatin (oxaliplatin) has been approved by the FDA. It is unlikely to increase the drug's patient base, but the new formulation should consolidate Sanofi-Aventis's position as one of the major players in colorectal cancer treatment.

Shire: Adderall should recover from Canadian blow

Shares in the UK's third-biggest drug manufacturer Shire slumped on news of Adderall XR's withdrawal in Canada, prompted after Shire provided information about 20 sudden deaths in patients taking Adderall XR for ADHD. The withdrawal from Canada will cause even bigger problems for Shire if the FDA chooses to follow Health Canada's lead, but at present this seems unlikely.

GenVec: getting the green light

Following submission of safety data from a Phase II clinical trial to the FDA, clearance has been granted for GenVec [GNVC] to resume clinical development of TNFerade in pancreatic adenocarcinoma patients. This is good news both for the company and for patients, since TNFerade holds real promise at this early stage for this notoriously hard-to-treat form of cancer.

Biogen-IDEC / Elan: Tysabri affirms its potential

Biogen-IDEC [BIIB] and Elan [ELA.L] have announced that Tysabri has demonstrated significant improvements in disability progression and reduction in the rate of relapse in relapse-remitting multiple sclerosis (RRMS). The outlook for the drug is therefore extremely bright, and it could cannibalize revenues of rival RRMS products significantly. Tysabri may yet herald a new dawn in RRMS treatment.

Elan: easing pain in the EU

The European approval of Prialt (ziconotide) is the first for an intrathecal chronic pain drug for many years, and Prialt should now provide an effective and safe treatment for patients for whom other treatment has failed. However, given the limited number of patients for whom Prialt treatment will be suitable, the drug has limited market potential.

Testing times for Tysabri

Biogen Idec and Elan have announced the voluntary suspension from sale and withdrawal from clinical trials of Tysabri, the companies' novel therapy for the treatment of relapse remitting multiple sclerosis (RRMS). This move has already slashed share prices of both companies. Even if Tysabri is relaunched with some form of black-box warning, doubts will surely linger over its long-term safety.

Big pharma set to move into HIV/hepatitis co-infection niche

While the European Consensus Conference gained a consensus agreement for the treatment of HIV/HCV coinfection, the definition of best practice for the management of HIV/HBV remains clouded by the lack of data. However, this is certain to change with BMS' entecavir trial in HIV/HBV co-infected patients. Datamonitor's Brigitte de Lima reports...

Renovis: all is not lost

Renovis will be disappointed at the termination of a Phase II trial in PHN for its novel neuropathic pain therapy REN-1654. However, it should not give up hope: it is rightly planning to continue trials of the drug in sciatica, which could yet offer the access to the broader pain market. If Renovis can prove efficacy a marketing partnership for REN-1654 would be a logical next step for the firm.
